Associations between maternal matrix metalloproteinase gene polymorphisms and birth weight

Objective. To investigate the associations between maternal matrix metalloproteinase gene polymorphisms and birth weight.
Materials and methods. In a cohort of pregnant women (n=317), single-nucleotide polymorphisms (SNPs) in five genes encoding matrix metalloproteinases (MMPs) were investigated: rs1799750 [1G>2G] in MMP-1, rs243865 [C>T] in MMP-2, rs3025058 [6A>5A] in MMP-3, rs11568818 [T>C] in MMP-7, and rs17577 [G>A] in MMP-9.
Results. Analysis of the associations between SNPs and birth weight identified two loci associated with this phenotype: the 5A allele of rs3025058 in MMP-3 [allelic model (β=-0.112, p=0.002, pperm=0.003), additive model (β=-0.125, p=0.001, pperm=0.001), dominant model (β=0.155, p=0.009, pperm=0.013), and recessive model (β=-0.194, p=0.002, pperm=0.002)] and the 2G allele of rs1799750 in MMP-1 [recessive model (β=0.153, p=0.015, pperm=0.018)]. The polymorphic loci associated with birth weight exert functional effects in various organs and tissues involved in determining birth weight. The 2G allele of rs1799750 in MMP-1 is associated with lower MMP-1 gene expression in fibroblast cell cultures and in the thyroid gland, lungs, and visceral and subcutaneous adipose tissue. In addition, 35 SNPs in linkage disequilibrium with rs3025058 in MMP-3 (r2>0.8) also affect MMP-1 gene transcription in the thyroid gland, visceral adipose tissue, and fibroblast cell cultures.
Conclusion. Thus, the 5A allele of rs3025058 in MMP-3 is associated with lower birth weight, whereas the 2G allele of rs1799750 in MMP-1 is associated with higher birth weight.
